Differentiated HepaRG cells - HPR116

HPR116 are HepaRG™ cells cryopreserved after differentiation

HPR116 is available through a Limited Use License Agreement.

After thawing, the differentiated HepaRG™ cells can be used immediately in suspension for clearance, metabolism or transporter studies.
They can also be seeded and used as little as 4 hours later for metabolism studies in monolayer.
A few days after seeding and culture, the cells will form a co-culture of hepatocytes and of biliary-like epithelial cells. They can then be used for induction, toxicity or metabolism studies.

The cells are provided in a 1 ml vial with 8 x 106 viable cells.
Cell seeding:
- One vial will seed one 96-well plate (72 000 cells/well)
- One vial will seed 16 wells of one 24-well plate (480 000 cells/well)
Please carefully respect recommended cell seeding density.

1 mL / 8 million cells / 10% DMSO

Thawing process
(e.g: 1 vial for plating into 24 MW-plate)

1. Thaw and dilute cells with 9 mL of thawing medium (10 ml / 8 million cells)
2. Centrifuge, wash and re-suspend cells with 5 mL thawing / plating medium
3. Count cell viability, concentration and yield   
4. Adjust cell concentration to 0.96 million cells/mL  and plate cells onto 24 MW-plate  (0.5 mL / well)

Quality controls include

  • Number of viable cells per vial
  • Post-thaw viability
  • Cell confluence 6 hours after thawing and seeding cells ≥85%
  • Cell morphology
  • Mycoplasma detection (negative)
  • Microbial sterility
  • Enzymatic activities
  • Inducibility of CYP1A2, CYP3A4/5, CYP2B6 at day 3 (48 hours induction)
  • Uptake transporter activities

Storage and delivery conditions for HPR116 cells

Differentiated HepaRG™ cryopreserved cells are delivered in dry ice or in a nitrogen vapour dewar. If you receive them in dry ice, ensure shipment time did not exceed 48 hours.
HPR116 cells can be stored for 5 years in liquid nitrogen.

HPR116 – Cells, plates, media and supplements

  • HPR116-TA08 - Cryopreserved HepaRG™ cells
    1 mL in vial with ≥8 x 106 viable cells. One vial will fully seed one 96-well plate (72,000 cells/well) and 3 vials will fully seed one 24-well plate (480,000 cells/well)
The following supplements are available from Biopredic International:
  • MIL600 - Basal hepatic cell medium 100 ml
  • ADD620 - HepaRG™ Maintenance/Metabolism Medium Supplement with antibiotics
    15.3 mL. When combined with 100 ml of base medium, sufficient for 12 medium changes for studies where prolonged cell life or enhanced metabolic function is required
  • ADD640 - HepaRG™ Induction Medium Supplement with antibiotics
    3.7 mL. When combined with 100 ml of base medium, sufficient for 10 medium changes for induction studies
  • ADD650 - HepaRG™ Serum-free Induction Medium Supplement with antibiotics
    1.6 mL. When combined with 100 ml of base medium, sufficient for 10 medium changes for induction studies
  • ADD670 - HepaRG™ Thawing/Plating/General Purpose Medium Supplement with antibiotics
    13.8 mL. When combined with 100 ml of base medium, sufficient to thaw 4 vials
